TheEZH2(A677G)ChemiluminescentAssayKitisdesignedtomeasureactivityofthemutantEZH2complex[EZH2(A677G)/EED/SUZ12/RbAp48/AEBP]forscreeningandprofilingpurposes.ThekeytotheEZH2(A677G)AssayKitisahighlyspecificantibodythatrecognizesmethylatedHistoneH3K27.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,S-adenosylmethionineisincubatedwithasamplecontainingassaybufferandmethyltransferaseenzymeforonehour.Next,primaryantibodyisadded.Finally,thestripsaretreatedwithanHRP-labeledsecondaryantibodyfollowedbyadditionoftheHRPsubstratetoproducechemiluminescencethatcanthenbemeasuredusingachemiluminescencereader.
