TheCBX1InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofCBX1bindingtoitssubstrate.ThekitcomesinaconvenientAlphaLISA®format,withenoughbiotinylatedpeptidesubstrate,assaybuffer,detectionbufferandpurifiedGST-taggedCBX1chromodomaintoperformatotalof384enzymereactions.ThekeytothekitisthespecificbindingoftheCBX1chromodomaintothemethylated-peptidesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingCBX1andaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforonehour.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofpotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
