Alvespimycinhydrochlorideisthehydrochloridesaltofalvespimycin,ananalogueoftheantineoplasticbenzoquinoneantibioticgeldanamycin.AlvespimycinbindstoHSP90,achaperoneproteinthataidsintheassembly,maturationandfoldingofproteins.Subsequently,thefunctionofHsp90isinhibited,leADIngtothedegradationanddepletionofitsclientproteinssuchaskinasesandtranscriptionfactorsinvolvedwithcellcycleregulationandsignaltransduction.
Description:
AlvespimycinHCl,alsoknownas17-DMAG,isapotent,water-solubleHSP90inhibitor.Alvespimycinhasdiverseanti-tumoractionsandhaspotentialintreatinganumberofdifferenttypesofcancers,includingovarian,prostate,melatnoma,non-small-celllungcancer,andseveralleukemias(chroniclymphocytic,acutemyeloid).ThiscompoundalsosuppressesinflammationbyinterferingwithsignalingthroughtheNF-κBpathway.
