TheJHDM1DHomogeneousAssayKitisdesignedtomeasureactivityofJHDM1Dforscreeningandprofilingapplications.JHDM1D,alsoknownasKDM7AandKIAA1718,isaJumonjiC(JmjC)domain-containinghistonelysinedemethylasethatexhibitsdemethylationactivitytowarddimethyl-lysine9anddimethyl-lysine27onhistoneH3(H3K9me2andH3K27me2respectively).ThekeytotheJHDM1DHomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,asamplecontainingJHDM1Denzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).ThepresenceofculturemediumRPMI1640at>1%leadstosignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
