Description:
TheFBXL11HomogeneousAssayKitisdesignedtomeasuretheactivityoftheFBXL11forscreeningandprofilingapplications.ThekeytotheFBXL11HomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,asamplecontainingFBXL11enzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

SuppliedAs:
TheFBXL11HomogeneousAssayKitcomesinaconvenientAlphaLISA®format,withbiotinylatedhistoneH3peptidesubstrate,primaryantibody,demethylaseassaybuffer,andpurifiedFBXL11for384enzymereactions.
Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).ThepresenceofculturemediumRPMI1640at>1%leadstosignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
