Description:
TheFBXL10HomogeneousAssayKitisdesignedtomeasuretheactivityoftheFBXL10forscreeningandprofilingapplications.ThekeytotheFBXL10HomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,asamplecontainingFBXL10enzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).ThepresenceofculturemediumRPMI1640at>1%leadstosignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
