TheFluorogenicSIRT5AssayKitisacompleteassaysystemdesignedtomeasureSIRT5activityforscreeningandprofilingapplications.TheFluorogenicSIRT5AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlythreesimplestepsonamicrotiterplateareneededtoanalyzetheSIRT5activitylevel.First,theSirtuin5fluorometricsubstrate(SIRTsubstrate5),containingasuccinylatedlysinesidechain,isincubatedwithpurifiedSIRT5enzyme.ThedesuccinylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
