TheFluorogenicSIRT3AssayKitisacompleteassaysystemdesignedtomeasureSirtuin3(SIRT3)activityforscreeningandprofilingapplications.TheFluorogenicSIRT3AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheSIRT3activitylevel.First,theHDACfluorometricsubstrate(HDACsubstrate1),containinganacetylatedlysinesidechain,isincubatedwithpurifiedSIRT3enzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
