TheFluorogenicSIRT1AssayKitisacompleteassaysystemdesignedtomeasureSirtuin1(SIRT1)activityforscreeningandprofilingapplications.TheFluorogenicSIRT1AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheSIRT1activitylevel.First,theSIRTfluorometricsubstrate(SIRTsubstrate1),containinganacetylatedlysinesidechain,isincubatedwithpurifiedSIRT1enzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
