Background:
HDACsregulatecellularprocessesbycatalyzingthehydrolysisofanacetylgroupfromacetyllysinesinmodifiedproteins.IntheHDACassay,fluorescent-dyemoleculesareattachedtoapeptidecontainingacetyllysine.Attachmenttothepeptidequenchesthefluorescenceofthedye.AftertreatmentofthepeptidewithanHDAC,thereactionismixedwithadevelopmentsolutionthatisspecificfornonacetylatedlysines.IftheacetylgrouphasbeenremovedfromthelysinebytheHDAC,thissolutionwillreleasethedyeallowingforfluorescence.FluorescenceisthereforedirectlyrelatedtoHDACactivity.
Description:
TheFluorogenicHDACAssayKitisacompleteassaysystemdesignedtomeasurehistonedeacetylase(HDAC)class1activityforscreeningandprofilingapplications.Thekitcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentHDACactivitymeasurements.Inaddition,thekitincludespurifiedHDAC2enzymeandapotentHDACinhibitor,TrichostatinA,foruseasapositiveandnegativecontrol.TheFluorogenicHDACAssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheHDACactivitylevel.First,theHDACfluorometricsubstrate,containinganacetylatedlysinesidechain,isincubatedwithpurifiedHDACenzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereaderat485nm(excitation)/528nm(emission).OurotherHDACkit(#50033)containsasubstratethatisexcitedatwavelengths350-380nmandfluorescesatwavelengths440-460nm.However,thesubstrateinthiskitfluorescesatlonger(green)wavelengths:485nm(excitation)/528nm(emission).ItismostusefulwhenthesampleorinhibitorfluorescesatwavelengthsthatoverlapthoseofourotherHDACsubstrate
