Background:
HDACsregulatecellularprocessesbycatalyzingthehydrolysisofanacetylgroupfromacetyllysinesinmodifiedproteins.IntheHDACassay,fluorescent-dyemoleculesareattachedtoapeptidecontainingacetyllysine.Attachmenttothepeptidequenchesthefluorescenceofthedye.AftertreatmentofthepeptidewithanHDAC,thereactionismixedwithadevelopmentsolutionthatisspecificfornonacetylatedlysines.IftheacetylgrouphasbeenremovedfromthelysinebytheHDAC,thissolutionwillreleasethedyeallowingforfluorescence.FluorescenceisthereforedirectlyrelatedtoHDACactivity.
Description:
TheFluorogenicHDACClass2aAssayKitisacompleteassaysystemdesignedtomeasurehistonedeacetylase(HDAC)class2aactivityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentHDACactivitymeasurements.Inaddition,thekitincludespurifiedHDAC4andapotentHDACinhibitor,TrichostatinA,foruseasapositiveandnegativecontrol.TheFluorogenicHDACClass2aAssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheHDACclass2aactivitylevel.First,theHDACfluorometricsubstrateisincubatedwithpurifiedHDAC4enzyme.ThedeacetylationsensitizesthesubstratesosubsequenttreatmentwiththeLysineDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
