Description:
TheMycPathwayReporterkitisdesignedformonitoringtheactivityoftheMycsignalingpathwayinculturedcells.Thekitcontainsatransfection-readyexpressionvectorforc-MycandMycluciferasereportervector.Insidethecells,c-MycwillbindtoMax,translocatetothenucleus,andinduceexpressionoftheMycluciferasereportervector.ThisreportercontainsthefireflyluciferasegeneunderthecontrolofmultimerizedMycresponsiveelementslocatedupstreamofaminimalpromoter.TheMycreporterispremixedwithconstitutively-expressingRenilla(seapansy)luciferasevector,whichservesasaninternalpositivecontrolfortransfectionefficiency.
Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwithconstitutivelyexpressingRenillaluciferasevectorasanegativecontrol.Thenon-inducibleluciferasevectorcontainsafireflyluciferasegeneunderthecontrolofaminimalpromoter,butwithoutanyadditionalresponseelements.Thenegativecontroliscriticalfordeterminingpathway-specificeffectsandbackgroundluciferaseactivity.
