ThePI3K/AKTsignalingpathwayisessentialforcellgrowthandsurvival.Disruptionofthispathwayoritsregulationhasbeenlinkedtoavarietyofcancersandcoronarydiseases.MammalianFOXOprotein(FOXO1,FOXO3,FOXO4),asubgroupofForkheadtranscriptionfactors,isamongthebestcharacterizedtargetsofthePI3K/AKTsignalingpathway.Thesetranscriptionfactorsfunctionasatriggerforapoptosisbyupregulatinggenesnecessaryforcelldeath.InsulinorgrowthfactorsinduceactivationofPI3K,whichinturnactivatesAKT.AKTdirectlyphosphorylatesFOXOs,resultingintheexportofFOXOsfromthenucleustothecytoplasm,therebyinhibitingFOXO-dependenttranscription.

TheFOXOReporterkitisdesignedtomonitoractivityofthePI3K/AKTsignalingpathwayandthetranscriptionalactivityofFOXOproteinsinculturedcells.Thekitcontainsthetransfection-readyFOXO3expressionvectorandtheFOXOluciferasereportervector,whichisaPI3K/Aktpathway-responsivereporter.ThisreportercontainsafireflyluciferasegeneunderthecontrolofmultimersoftheFOXOresponsiveelementlocatedupstreamofaminimalpromoter.TheFOXOreporterispremixedwithaconstitutively-expressingRenilla(seapansy)luciferasevector,whichservesasaninternalcontrolforthetransfectionefficiency.Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwithconstitutively-expressingRenillaluciferasevectorasnegativecontrol.Thenon-inducibleluciferasevectoralsocontainsthefireflyluciferasegeneunderthecontrolofaminimalpromoter,butwithoutanyadditionalresponseelements.Thenegativecontroliscriticaltodeterminingpathway-specificeffectsandbackgroundluciferaseactivity.
