TheCRE/CREBReporterkitisdesignedformonitoringtheactivityofthecAMP/PKAsignalingpathwayinculturedcells.Thekitcontainstransfection-readyCREluciferasereporter.ThisreportercontainsthefireflyluciferasegeneunderthecontrolofmultimerizedcAMPresponseelement(CRE)locatedupstreamofaminimalpromoter.ElevationoftheintracellularcAMPlevelactivatescAMPresponseelementbindingprotein(CREB)tobindCREandinducestheexpressionofluciferase.TheCREreporterispremixedwithconstitutively-expressingRenilla(seapansy)luciferasevectorthatservesasaninternalcontrolfortransfectionefficiency.Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwiththeconstitutivelyexpressingRenillaluciferasevectorasanegativecontrol.Thenon-inducibleluciferasevectorcontainsthefireflyluciferasegeneunderthecontrolofaminimalpromoter,butwithoutanyadditionalresponseelements.Thenegativecontroliscriticaltodeterminingpathway-specificeffectsandbackgroundluciferaseactivity.
