TheNrf2antioxidantresponsepathwayplaysanimportantroleinthecellularantioxidantdefense.Nrf2,abasicleucinezippertranscriptionfactor,inducestheexpressionofantioxidantandphaseIIenzymesbybindingtotheARE(antioxidantresponseelement)regionofthegenepromoter.Underbasalconditions,Nrf2isretainedinthecytosolbybindingtothecytoskeletalproteinKeap1.UponexposuretooxidativestressandotherAREactivators,Nrf2isreleasedfromKeap1andtranslocatestonucleus,whereitcanbindtotheARE,leADIngtotheexpressionofantioxidantandphaseIIenzymesthatprotectthecellfromoxidativedamage.
Description:
TheAREReporterkitisdesignedformonitoringtheactivityoftheNrf2antioxidantpathwayinculturedcells.Thekitcontainsatransfection-readyAREluciferasereportervector,whichisanNrf2pathway-responsivereporter.ThisreportercontainsafireflyluciferasegeneunderthecontrolofmultimerizedAREresponsiveelementslocatedupstreamofaminimalpromoter.TheAREreporterispremixedwithaconstitutivelyexpressingRenilla(seapansy)luciferasevectorthatservesasaninternalcontrolfortransfectionefficiency.Thekitalsoincludesanon-inducIBLefireflyluciferasevectorpremixedwithconstitutivelyexpressingRenillaluciferasevectorasnegativecontrol.Thenon-inducibleluciferasevectorcontainsafireflyluciferasegeneunderthecontrolofaminimalpromoter,withoutanyadditionalresponseelements.Thisnegativecontroliscriticaltodeterminingpathway-specificeffectsandbackgroundluciferaseactivity.
