TheBRD4(BD1)InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofBRD4bromodomain1(BD1)frombindingtoitssubstrate.ThekeytotheBRD4(BD1)InhibitorScreeningAssayKitisthehighlyspecificbindingoftheBRD4bromodomain1totheacetylatedhistonesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingBRD4bromodomain1andaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

KitcomesinaconvenientAlphaLISA®format,withbiotinylatedhistonepeptidesubstrate,assaybuffer,detectionbufferandpurified,GST-taggedBRD4BD1toperformatotalof384enzymereactions.
Contraindications:
DMSOabove0.5%.OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptBRD-ligandinteraction.AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
