TheBRD3(BD1)InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofBRD3bromodomain1(BD1)frombindingtoitssubstrate.ThekeytotheBRD3(BD1)InhibitorScreeningAssayKitisthehighlyspecificbindingoftheBRD3bromodomain1totheacetylatedhistonesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingBRD3bromodomain1andaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

KitcomesinaconvenientAlphaLISA^®format,withbiotinylatedhistonepeptidesubstrate,assaybuffer,detectionbufferandpurified,GST-taggedBRD3BD1toperformatotalof384enzymereactions.

Contraindications:

DMSOabove0.5%.OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptBRD-ligandinteraction.AvoidgreenandbluedyesthatabsorblightintheAlphascreen^®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN₃)ormetalions(Fe²⁺,Fe³⁺,Cu²⁺,Zn²⁺andNi²⁺).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen^®assays.
