TheBAZ2BInhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofBAZ2Bbindingtoitssubstrate.ThekeytotheBAZ2BInhibitorScreeningAssayKitisthehighlyspecificbindingoftheBAZ2Bbromodomaintotheacetylatedhistonesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingBAZ2Bandaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.
SuppliedAs:
KitcomesinaconvenientAlphaLISA®format,withbiotinylatedhistonepeptidesubstrate,assaybuffer,detectionbufferandpurifiedHis-BAZ2Bbromodomaintoperformatotalof384enzymereactions.
Contraindications:
DMSOabove0.5%.OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptBRD-ligandinteraction.AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
