ApoptosisActivator2isacell-permeableindoledionecompoundthatactivatescaspasesinacytochromec-dependentmannerandinducesapoptosisintumorcellsbypromotingtheoligomerizationofApaf-1intothematureapoptosome.Itscytotoxicityisselectivetowardstumorcells(IC50<10µMforJurkat,Molt-4,andCCRF-CEM).ApoptosisActivator2showslittleornoeffectonnon-tumorcellsorcellsthataredeficientincaspase-3orApaf-1(IC50>40µM).Throughtheinductionofapoptosis,ApoptosisActivator2notablyinducescaspase-3activation,PARPcleavageandDNAfragmentation.Moreover,thisinductionofapoptosisonlyoccursintumorcelllines(suchasbreast,lung,colon,andepidermalcancercelllines).
