Thep300HomogeneousAssayKitcontainsanAlphaLISA®assaydesignedforscreeninginhibitorsofp300inaconvenient384-wellformat.Thisefficientmethodrequiresnotime-consumingwashingsteps.Onlyafewsimplestepsonamicrotiterplateareneeded.TheassayisbasedontheenzymatictransferofanacetylgroupfromacetylCoAtoaspecificlysineresiduewithinabiotinylatedpeptidesubstrate.Afterthep300reaction,thesampleisincubatedwithacombinationofanantibodythatspecificallybindstheacetylatedpeptideandacceptorbeadsthatbindtheantibody.Thisisfollowedbyincubationwithstreptavidin-labeleddonorbeadsandreADIngoftheAlpha-counts.
